Kazia Therapeutics ($KZIA) is a catalyst-driven setup centered on upcoming ASCO data from the ongoing Phase 1b ABC-Pax study evaluating paxalisib in metastatic triple-negative breast cancer (TNBC). The update is expected to include additional efficacy, safety, and circulating tumor cell (CTC) biomarker data from patients treated with paxalisib in combination with pembrolizumab and chemotherapy, following earlier disclosures showing >50% reductions in CTCs and CTC clusters alongside early partial responses and a complete metabolic response under expanded access.
The key question is whether those early signals mature into clinically credible efficacy in a setting where prior PI3K inhibitors have largely failed due to toxicity, weak durability, and limited differentiation. Investors will focus on confirmed response rates, durability, visceral metastasis control, and whether the translational biomarker data correlates with actual tumor regression rather than remaining primarily mechanistic.
The broader debate is whether paxalisib is genuinely differentiated from prior PI3K pathway failures or simply being reframed through newer epigenetic and immunotherapy language. The company is increasingly positioning paxalisib as an “immune reprogramming” backbone capable of reversing immune exhaustion and improving checkpoint sensitivity in resistant tumors. If ASCO shows deeper responses, manageable tolerability, and continued consistency between biomarker shifts and clinical outcomes, bulls will argue the asset has broader solid tumor potential beyond CNS disease. If efficacy remains anecdotal or heavily dependent on translational framing, the story risks reverting back to another mechanistically interesting but clinically unproven PI3K program.
The setup remains highly binary because positive ASCO data could materially improve partnering leverage and platform credibility, while ambiguous or incremental results would likely reinforce concerns that the narrative is advancing faster than the underlying clinical evidence.
Options are pricing a highly binary event with roughly ±50% expected movement into expiration, while the broader chain still reflects tail scenarios approaching +100% upside if ASCO materially rerates the paxalisib narrative.
Every Monday we are out with public biotech research, profiling biotech companies with near term catalysts. This will focus primarily on the major market moving biotech events such as data readouts from the major scientific conferences as well as potential regulatory approvals.
We will not offer specific trading advice (and we do not hold any publicly traded biotech equity), but rather flag emerging events that will likely materially drive stock prices (sometimes +/-100% in either direction).

HOW TO PLAY BIOTECH APPROVAL/DATA READOUTS ANNOUNCEMENTS
While biotech equities will move on news of announcement, often if there is high conviction in a positive read, a large run up ahead of the news will occur and the actual event turns into a sell the news event. Sometimes, investors wait for the actual event to make a long/short decision. And obviously if the news is disappointing (more often than not) the stock will tank.
This is not financial advice so you will have to make your own call on how to best play these events. Our aim is simply to flag catalysts with high projected volatility.
