This week, we’re digging into the return of the mega-blockbuster and asking whether the next generation of oncology drugs could produce another Keytruda-sized franchise. Using the RAS inhibitor story as the jumping-off point, we break down what it actually takes to build a $20 billion drug, why moving into earlier lines of treatment can dramatically change the commercial opportunity, and where seemingly reasonable peak-sales models can quickly turn into fantasy. We also look beyond RAS at ADCs, radiopharma and bispecifics, and ask the question that tends to get lost when the TAM starts looking enormous: does a massive therapeutic category create one dominant drug, or simply attract enough competition to split the economics across five or six of them?
